Oxygen-sensitive ccdA antitoxin expression system
BBa_K4256155 is an expression system of the antitoxin protein ccdA (BBa_K4256111) under the control of the oxygen-controlled promoter HIP-1 (BBa_K4256100). Under anaerobic conditions,ccdA is normally expressed and inhibits the killing effect of ccdB; under normoxic conditions ccdA expression is repressed.Under hypoxic conditions, the stable presence of the [4Fe-4S] cluster in the FNR protein drives the FNR protein to form a homodimer, and this dimer can induce the initiation of downstream ccdA expression; under normoxic conditions, the [4Fe-4S] cluster is disassembled, and the FNR dimer is depolymerized, so the initiation of ccdA transcription is inhibited.
